I agree that the first thing you should do is sign up for their player's club card. That gets you earning comps right away, including discounts for rooms and even free nights in combination with paid rooms.
Then, always use the card whenever you are gaming, whether inserting it in the slots, or handing it to the dealer at the table games. The bigger the bet and the longer you play, the faster you will earn comp "points." These points can be applied to rooms, food, shopping and other uses. The cards also get you email and snail mail offers, pretty much every month, including invitation to card members only events like tournaments, shows as well as free or discount room offers.

sign up for the players club and spend a LOT of money. comps aren't easy to get anymore unless you are a big spender. last time i went to vegas, i stayed at the same casino i always stay at and gambled about $500...i ended up with like $5 worth of comp money at the end...the way it works is that you use your Player's Club card at all of the machines and tables that you are gambling on...at the end of your trip, you go to the Players Club area and they check your card to see if you earned anything...once, we spent like a week there and i earned enough to buy a pair of sunglasses and some dice with my name on them in the casino store...that's been about the extent of it.
basically, comps are given because you are spending so much with the casino already that things like dinners and rooms are nothing comparitively...maybe 1% of people that go to vegas actually spend that much.
other than spending a ton, you really only have a decent chance if you go to the hotel extremely frequently [like people that play in slot and poker tournaments all the time] or you win some sort of big jackpot that you then spend at the casino.
now if you're looking for discounts and coupons, those are EVERYWHERE...definitely join the Player's Club, and look on the internet, and you can get crazy discounts while your there
ADDED: the person above is correct...you can easily get comped rooms at Indian Casinos, and places outside of Vegas like Reno, but comps in Vegas itself are pretty much nonexistent to the average person
